目的 :探讨运动对骨细胞凋亡以及IGF -1表达变化的影响并讨论其在骨代谢变化中的可能意义。方法 :3 0只 5月龄雄性SD大鼠均分为运动组和正常对照组。运动组大鼠在跑台上进行中等强度运动 8周。8周后处死动物 ,对血钙、血碱性磷酸酶、血骨钙素、尿钙、尿肌苷进行检测。采用双能X线骨密度仪对各组大鼠股骨骨密度进行测定 ;应用原位凋亡法计数股骨髁部松质骨骨细胞凋亡指数 ;对股骨髁部进行IGF-1免疫组织化学染色 ,并对表达强度进行评定。结果 :运动组大鼠血碱性磷酸酶、血骨钙素显著高于对照组(P <0 0 5 ,P <0 0 1 ) ;血钙、尿钙 /尿肌苷两组之间无统计学差异。运动组大鼠股骨骨密度明显高于对照组 (P <0 0 5 )。运动组大鼠小梁骨中骨细胞凋亡比率显著低于对照组 (P <0 0 5 )。运动组大鼠IGF -1阳性骨细胞表达强度显著高于对照组 (P<0 0 1 )。结论 :运动抑制骨细胞凋亡同时促进骨组织局部细胞因子IGF -1的表达。
OBJECTIVE: To investigate the effect of exercise on the apoptosis of osteoblasts and the expression of IGF-1 and its possible significance in the changes of bone metabolism. Methods: Thirty male SD rats of 5 months old were divided into exercise group and normal control group. Exercise rats in the treadmill for moderate intensity exercise for 8 weeks. After 8 weeks, the animals were sacrificed and the blood calcium, blood alkaline phosphatase, blood osteocalcin, urinary calcium and urinary creatinine were tested. The BMD of the femur in each group was measured by dual-energy X-ray absorptiometry. The apoptosis index of cancellous bone in the femoral condyle was measured by in situ apoptosis method. IGF-1 immunohistochemical staining , And evaluate the intensity of expression. Results: The blood alkaline phosphatase and serum osteocalcin in exercise group were significantly higher than those in control group (P <0.05, P <0.01). There was no statistical significance between the two groups Differences The BMD of femur in exercise group was significantly higher than that in control group (P <0 05). The ratio of apoptosis of trabecular bone in exercise group was significantly lower than that in control group (P <0.05). The expression of IGF-1 positive cells in exercise group was significantly higher than that in control group (P <0.01). Conclusion: Exercise inhibits apoptosis of osteocytes and promotes the expression of local cytokine IGF - 1 in bone tissue.
