目的 研究从病人和水产品中分离到的 5株O139群霍乱弧菌的生物学性状。方法 参照《霍乱防治手册》对 5株菌的生长特征、生化特征、血清学反应及药物敏感性进行鉴定 ;应用多重PCR方法对所试菌株进行毒力相关基因检测 ,用Vero细胞测定方法检测ctxA阳性菌株霍乱肠毒素的表达。结果 5株O139群霍乱弧菌生物学性状和血清学反应与阳性对照菌体O139群霍乱弧菌MO4 5基本相同 ;2株来自重型病人和 1株来自水产品的菌株ctxA、zot、ace、tcpA、tcpI、hlyA、rtxA、rtxC、ompU、toxR 11种毒力相关基因全部阳性 ,经Vero细胞测定 ,其霍乱肠毒素效价达到 1∶16 0 ;在 2株来自轻型病人的菌株中仅hlyA、rtxA、rtxC、toxR 4种毒力相关基因为阳性。 结论 O139群霍乱弧菌致病 (泻 )的决定基因簇并非仅为ctxA、zot、ace、cep等毒力相关基因。
Objective To study the biological characteristics of five strains of Vibrio cholerae O139 isolated from patients and aquatic products. Methods The growth characteristics, biochemical characteristics, serological response and drug susceptibility of five strains were identified according to the Cholera Prevention and Treatment Manual. The virulence-related genes of the tested strains were detected by multiplex PCR and the ctxA Positive strains of cholera enterotoxin expression. Results The biological characteristics and serological responses of 5 strains of Vibrio cholerae O139 were basically the same as that of V. cholerae O139 strain O139. Two strains from heavy-duty patients and one from ctxA, zot, ace and tcpA , TcpI, hlyA, rtxA, rtxC, ompU, toxR all virulence-related genes were all positive, as determined by Vero cells, its cholera enterotoxin titer reached 1: 160; in two strains from light patients only hlyA, rtxA, rtxC, toxR 4 virulence-associated genes were positive. Conclusion The gene cluster of pathogenic Vibrio cholerae O139 (diarrhea) is not only related to virulence genes such as ctxA, zot, ace and cep.
