目的探讨血小板反应素(TSP)mRNA表达、血浆血小板第4因子(PF4)水平与甲状腺乳头状癌(PTC)颈部淋巴结转移的关系。方法应用逆转录多聚酶链反应(RT-PCR)技术检测40例PTC癌组织和10例癌旁正常组织TSP-1、TSP-2 mRNA的表达,并用酶标记免疫分析法(ELISA)测定40例PTC患者、20例健康体检者(对照组)的血浆PF4水平。结果(1)PTC癌组织TSP-1 mRNA、TSP-2 mRNA阳性表达率分别为45.0%、47.5%,它们均明显低于正常组织(80.0%)(P<0.01),而且TSP-1 mRNA、TSP-2 mRAN阳性表达PTC组的颈部淋巴结转移率均明显低于TSP-1 mRNA、TSP-2 mRNA阴性表达PTC组(33.3% vs 90.9%、42.1% vs 85.7%,P<0.01)。(2)正常对照组、无淋巴结转移的PTC组和伴淋巴结转移的PTC组间的PF4血浆浓度呈线性递减趋势,其中无淋巴结转移组的PF4血浆浓度(2.96±0.21)μg/L高于转移组(0.98±0.17)μg/L(P<0.05)。(3)TSP-1 mRNA、TSP-2 mRNA阳性表达PTC组的血浆PF4水平分别明显高于TSP-1 mRNA、TSP-2 mRNA阴性表达组[(2.36±0.91)μg/L vs(1.11±0.60)μg/L,(2.15±1.00)μg/L vs(1.23±0.72)μg/L](P<0.01)。结论缺乏TSP-1 mRNA、TSP-2 mRNA表达及血浆低水平的PF4诸因素可能为促进PTC发生颈部淋巴结转移起到了推波助澜的作用,TSP-1、TSP-2与PF4在抑制PTC颈部淋巴结转移中起着互相协同的作用。
Objective To investigate the relationship between the expression of thrombospondin (TSP) mRNA, the level of plasma platelet factor 4 (PF4) and cervical lymph node metastasis in papillary thyroid carcinoma (PTC). Methods The expressions of TSP-1 and TSP-2 mRNA in 40 cases of PTC and 10 cases of adjacent normal tissues were detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Enzyme-linked immunosorbent assay (ELISA) Patients, 20 healthy subjects (control group) plasma PF4 levels. Results The positive rates of TSP-1 mRNA and TSP-2 mRNA in PTC tissues were 45.0% and 47.5%, respectively, which were significantly lower than those in normal tissues (80.0%) (P <0.01) TSP-2 mRAN positive expression in PTC group was significantly lower than that in TSP-1 mRNA and TSP-2 mRNA negative group (33.3% vs 90.9%, 42.1% vs 85.7%, P <0.01). (2) The plasma concentration of PF4 in normal control group, PTC group without lymph node metastasis and PTC with lymph node metastasis showed a decreasing trend linearly. The plasma concentration of PF4 in non-lymph node metastasis group (2.96 ± 0.21) μg / L was higher than that in metastasis Group (0.98 ± 0.17) μg / L (P <0.05). (3) The levels of plasma PF4 in TSP-1 mRNA and TSP-2 mRNA positive group were significantly higher than those in TSP-1 mRNA and TSP-2 mRNA group [(2.36 ± 0.91) μg / L vs (1.11 ± 0.60 (2.15 ± 1.00) μg / L vs (1.23 ± 0.72) μg / L] (P <0.01). Conclusion The lack of TSP-1 mRNA, TSP-2 mRNA expression and low plasma PF4 may promote the occurrence of cervical lymph node metastases in PTC.
