目的通过超快速液相色谱-三重四极杆飞行时间串联质谱(UFLC-Triple TOF-MS/MS)技术对厚朴主要化学成分进行分析鉴定。方法采用Agilent ZORBAX SB-C_(18)色谱柱(4.6 mm×250 mm,5μm),以0.1%甲酸水溶液-乙腈为流动相梯度洗脱;质谱使用电喷雾(ESI)离子源,在正离子与负离子两种模式下采集数据。依据高分辨质谱提供的准分子离子峰和碎片离子的精确分子质量信息,结合标准品对照与参考相关文献数据,对厚朴进行成分鉴定。结果初步鉴定出24个化学成分,其中包括12个木脂素类、9个生物碱类和3个苯丙素苷类化合物。结论为进一步探究厚朴的药效物质基础和质量调控机制提供基础资料。
Objective To analyze and identify the chemical constituents of Magnolia officinalis by ultra fast liquid chromatography-triple quadrupole time-of-flight mass spectrometry (UFLC-Triple TOF-MS / MS) Methods The mobile phase was eluted with a mobile phase of 0.1% formic acid in acetonitrile on a ZORBAX SB-C 18 column (4.6 mm × 250 mm, 5 μm). The mass spectrometry was carried out using electrospray ionization (ESI) Negative ions two modes to collect data. Based on the accurate molecular mass information of quasi-molecular ion peak and fragment ion provided by high-resolution mass spectrometry, combined with the standard reference and related literature data, the constituents of Magnolia officinalis were identified. The results initially identified 24 chemical constituents, including 12 lignans, 9 alkaloids and 3 phenylpropanoid glycosides. Conclusion To provide basic information for further research on the material basis and quality control mechanism of Magnolia officinalis.
