本文用25%甘露醇注入兔的一侧颈内动脉,获得了满意的渗透性BBB开放模型,使静脉注射的EB在BBB开放侧脑半球着色。这种BBB开放是可逆 性的,约持续2小时,且不伴神经组织结构的损害。在BBB开放后的1、24、72小时,脑组织水分、Na~+Kt含量均在正常范围,与对照组比较无显著性差异(P>0.05)。超微结构观察的结果证实,渗透性BBB开放的形态学基础主要是脑毛细血管内皮细胞的皱缩,其间的紧密连接增宽。在BBB开放期间,由颈动脉或静脉应用MTX,能显著提高脑组织内的MTX浓度,前一途径可使BBB开放侧脑半球的药物浓度增至最高水平。本文研究结果提示:渗透性BBB开放是一种安全、简单可行的方法,为化疗药物入脑开辟了途径,有可能改善恶性脑瘤的化疗效果,对其它颅内疾病的治疗及某些基础学科的研究也具有意义。
In this study, we injected 25% mannitol into the carotid artery on one side of the rabbit and obtained a satisfactory open model of BBB permeability. Intravenous EBs were colored in the open hemisphere of the BBB. This BBB opening is reversible, about 2 hours, without damage to the nerve tissue structure. At 1, 24 and 72 hours after BBB was opened, brain water content and Na ~ + Kt content were in the normal range, no significant difference compared with the control group (P> 0.05). The results of the ultrastructural observation confirmed that the morphological basis of the openness of the osmotic BBB is mainly the collapse of the brain capillary endothelial cells with the broadening of tight junctions therebetween. During the BBB opening, the application of MTX from the carotid artery or vein can significantly increase the concentration of MTX in brain tissue. The former approach maximizes the drug concentration in the lateral hemispheres of the BBB. The results of this study suggest that the open BBB penetration is a safe, simple and feasible method for the chemotherapy drugs into the brain opened up ways that may improve the malignant brain tumor chemotherapy, the treatment of other intracranial diseases and some basic disciplines The research also makes sense.
