目的:观察绿原酸(chlorogenic acid)对血管生成的抑制作用及其机制。方法:采用鸡胚尿囊膜(CAM)模型观察绿原酸对血管生成的抑制作用。采用MTT、Transwell及Western blotting方法研究绿原酸抑制血管生成的机制。结果:100μg/ml绿原酸能明显抑制CAM新生血管的生长。绿原酸能抑制HUVEC细胞增长的IC50为38μmol/L;绿原酸浓度为25μg/ml、50μg/ml、100μg/ml时,HUVEC细胞迁移数和VEGF蛋白表达量均受到抑制,且呈剂量依赖性。结论:绿原酸能够抑制血管生成,该作用可能与下调血管内皮生长因子VEGF的表达有关。
Objective: To observe the inhibitory effect of chlorogenic acid on angiogenesis and its mechanism. Methods: The chick embryo allantoic membrane (CAM) model was used to observe the inhibitory effect of chlorogenic acid on angiogenesis. MTT, Transwell and Western blotting were used to study the mechanism of chlorogenic acid in inhibiting angiogenesis. Results: 100μg / ml chlorogenic acid can significantly inhibit the growth of CAM neovascularization. Chlorogenic acid inhibited the growth of HUVEC cells with an IC50 of 38μmol / L. When the concentrations of chlorogenic acid were 25μg / ml, 50μg / ml and 100μg / ml, the migration and VEGF protein expression were inhibited in a dose-dependent manner Sex. CONCLUSION: Chlorogenic acid can inhibit angiogenesis, which may be related to the downregulation of vascular endothelial growth factor VEGF expression.
