代谢组学是近年来发展起来的系统生物学分支,旨在揭示生物体受外界刺激后,体液、细胞及组织等生物样品中内源性代谢物的变化规律。气相色谱-质谱联用技术(GC-MS)具有较高的分辨率和检测灵敏度,并且配有标准谱图库,可以方便地鉴别代谢组分,因而在代谢组学中应用广泛。本文介绍了GC-MS代谢组学研究的关键技术,包括衍生化技术、代谢物鉴定及数据分析;同时,介绍了GC-MS用于检测小分子代谢物如氨基酸、脂肪酸、有机酸及糖类等的典型应用。
Metabonomics is a systematic biological branch developed in recent years to reveal the variation of endogenous metabolites in biological samples such as body fluids, cells and tissues after external stimuli. Gas chromatography - mass spectrometry (GC-MS) with high resolution and detection sensitivity, and with a standard spectral library, you can easily identify the metabolic components, and therefore widely used in metabolomics. This paper introduces the key technologies of GC-MS metabolomics research, including derivatization, metabolite identification and data analysis.
