试验在MS基本培养基上设0h15℃和16h25℃不添加生长调节剂,16h25℃添加多效唑(PP333)0.1mg/L,0.5mg/L和1.0mg/L共5个处理,研究各处理微型薯诱导及内源激素变化.结果表明:在低温黑暗(0h15℃)和培养基中添加PP333的高温长日(16h25℃)条件下,组培苗中赤霉素(GA3)质量分数在后期下降,能诱导微型薯形成;在16h25℃不添加生长调节剂的条件下,GA3质量分数较高,不能形成试管薯.在培养基中添加PP333可抑制组培苗GA3的合成,同时诱导试管薯形成,其微型薯数量和质量均优于0h15℃处理条件.对脱落酸(ABA)和茉莉酸(JA)的测定结果表明:无论试管薯形成与否,组培苗内ABA和JA的质量分数均随组培苗的生育进程呈上升趋势.因此,GA3下降是微型薯形成的必要条件,PP333可通过调节内源激素质量分数来逆转高温长日对块茎形成的抑制作用,ABA和JA与组培苗衰老的关系比与块茎形成的关系更为密切.

PP333 can reverse the inhibitory effect of high temperature and long day on tuber formation by regulating the content of endogenous hormones. The relationship between aging and tuber formation is more closely related.
