目的观察高血压患者血小板聚集功能变化是否与Bβ纤维蛋白原(Fib)基因启动子区-455G/A多态性有关。方法选取108例未经治疗的高血压患者,采用全血电阻抗法测定二磷酸腺苷(ADP)及花生四烯酸(AA)诱导的血小板聚集功能,并用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)方法测定BβFib基因启动子区-455G/A多态性,将其分为GG组和GA+AA组,比较两组血小板聚集功能变化。结果GA+AA组Fib浓度高于GG组(P<0.05),ADP诱导的血小板聚集能力GA+AA组低于GG组(P<0.05);而AA诱导的血小板聚集能力呈现GA+AA组高于GG组的趋势(P>0.05)。结论ADP诱导的血小板聚集功能与BβFib基因启动子区-455G/A多态性有关,GG基因型患者血小板聚集功能强于AA及GA基因型患者。但AA诱导的血小板聚集功能与其的关系呈现相反的趋势。
Objective To investigate whether changes in platelet aggregation in patients with hypertension are related to the -455G / A polymorphism in the promoter region of Bβ fibrinogen (Fib) gene. Methods 108 patients with untreated hypertension were enrolled in this study. The platelet aggregation function induced by adenosine diphosphate (ADP) and arachidonic acid (AA) was determined by whole blood impedance assay. The effect of polymerase chain reaction - restriction fragment length Polymorphism analysis (PCR-RFLP) method was used to determine the -455G / A polymorphism of BβFib promoter region, which was divided into GG group and GA + AA group. The platelet aggregation function was compared between the two groups. Results The concentration of Fib in GA + AA group was higher than that in GG group (P <0.05), ADP-induced platelet aggregation ability in GA + AA group was lower than that in GG group (P <0.05) In GG group (P> 0.05). Conclusion The platelet aggregation induced by ADP is related to the -455G / A polymorphism of BβFib gene promoter region. The GG genotype patients have stronger platelet aggregation than AA and GA genotypes. However, the relationship between AA-induced platelet aggregation and its function showed the opposite trend.
