目的 :通过免疫组化方法对视紫质 (rhodopsin)、胶质纤维酸性蛋白 (glialfibrillaryacidicprotein ,GFAP)、波形蛋白(vimentin)、血管内皮细胞生长因子 (vascularendothelialgrowthfactor,VEGF)的检测 ,从分子水平观察光动力疗法 (PDT)治疗后非脉络膜新生血管形成 (CNV)区的视网膜的蛋白表达情况 ,对PDT的安全性进行研究。方法 :采用过量的眼底激光破坏Bruch膜 ,建立猴眼CNV模型 ;将正常猴眼作为对照 1组 ,未作PDT治疗的CNV模型眼作为对照 2组 ;通过免疫组化的方法来研究PDT治疗后非CNV区 1、2、3、6个月的猴眼视网膜的视紫质抗体、胶质纤维酸性蛋白抗体、波形蛋白抗体、血管内皮细胞生长因子抗体及其原位杂交 ,检测PDT治疗组与对照组的视网膜抗体表达情况。结果 :猴眼视网膜非CNV区各层组织经PDT治疗后与对照组相比视紫质抗体、胶质纤维酸性蛋白抗体、波形蛋白抗体、血管内皮细胞生长因子抗体及血管内皮细胞生长因子原位杂交信号均未见明显差异。结论 :PDT治疗后并未引起视网膜各层蛋白表达异常 ,VEGFmRNA在分子水平亦未见表达改变 ,因此PDT治疗对视网膜组织是安全的。
OBJECTIVE: To detect the expressions of rhodopsin, glial fibrillary acid protein (GFAP), vimentin, and vascularendothelial growth factor (VEGF) by immunohistochemistry. Retinal protein expression in non-choroidal neovascularization (CNV) area after dynamic therapy (PDT) was studied, and the safety of PDT was studied. Methods: The Bruch’s membrane was destroyed by excessive fundus laser to establish the CNV model of monkey eyes. The normal monkey eyes were used as control group 1 and the CNV model eyes without PDT as control group 2, and the PDT group The retinal rhodopsin antibodies, glial fibrillary acidic protein antibodies, vimentin antibodies, vascular endothelial growth factor antibodies and their in situ hybridization in the non-CNV area for 1, 2, 3, Retinal antibody expression in the control group. Results: Compared with the control group, all layers of the non-CNV retina in the retina of the retina were treated with PDT, glial fibrillary acidic protein antibody, vimentin, VEGF and VEGF No significant differences in hybridization signals were observed. CONCLUSION: PDT does not cause any abnormal expression of protein in retina and VEGFmRNA has no change in molecular level. PDT treatment is safe to retinal tissue.
