应用高效液相色谱技术,测定头孢氨噻肟(CTX)及其主要代谢产物去鲸蜡基头孢氨噻肟(dCTX)对血液外淋巴液屏障的通透性。结果显示,CTX、dCTX均能通过血液外淋巴液屏障,dCTX在耳蜗外淋巴液中的浓度比CTX高1.17~3.81倍;静注CTX(100μg/kg)1h后,外淋巴液中CTX超过细菌性迷路炎常见致病菌最低抑菌浓度的10倍以上;注射6h后,外淋巴液中的CTX仍高于最低抑菌浓度。提示CTX可以作为临床用于细菌性迷路炎防治的有效药物。
High performance liquid chromatography (HPLC) was used to determine the permeability of cefotaxime (CTX) and its major metabolite cetylceramide (dCTX) to the extrahepatic lymph barrier. The results showed that both CTX and dCTX could pass the extra-blood lymph barrier, and the concentration of dCTX in cochlear extranodal fluid was 1.17 ~ 3.81 times higher than that of CTX. After intravenous injection of CTX (100μg / kg) for 1h, CTX more than bacterial vaginitis common pathogenic bacteria minimum inhibitory concentration of 10 times or more; injection 6h, CTX in the perilymph is still higher than the minimum inhibitory concentration. Tip CTX can be used as an effective clinical prevention and treatment of bacterial labyrinthine drugs.
