目的探讨压榨法提取的姜黄素对宫颈癌Hela细胞荷瘤裸鼠的治疗作用及其作用机制。方法将♀裸鼠腋下接种Hela细胞建立颈癌模型,随机均分为模型组、5-Fu组、姜黄素高、中、低剂量组(240、120、60 mg·kg-1);检测各组小鼠的凝血功能;免疫组化法检测微血管密度(MVD)及血管内皮生长因子(VEGF)、IFN-γ的表达。结果与模型组比较,姜黄素高、中、低剂量组的凝血时间显著延长(P<0.05),MVD计数显著降低(P<0.05),VEGF的表达降低(P<0.01),而IFN-γ的表达升高(P<0.05)。结论姜黄素可能通过调节VEGF、IFN-γ的表达来抑制新生血管的形成,改善裸鼠血液的高凝状态而治疗宫颈癌。
Objective To investigate the therapeutic effect and mechanism of curcumin extracted by pressing method on cervical cancer Hela cell-bearing nude mice. Methods Nude mice were inoculated with Hela cells into the nude mice to establish a model of cervical cancer. The models were randomly divided into model group, 5-Fu group, curcumin high, medium and low dose groups (240, 120, 60 mg · kg -1) The coagulation function of each group of mice was observed. The expression of MVD, VEGF and IFN-γ were detected by immunohistochemistry. Results Compared with the model group, the clotting time of curcumin in high, medium and low dose groups was significantly longer (P <0.05), the MVD was significantly lower (P <0.05) and the expression of VEGF was lower (P <0.01) (P <0.05). Conclusion Curcumin may treat cervical cancer by regulating the expression of VEGF and IFN-γ to inhibit the formation of new blood vessels and improve the hypercoagulability of blood in nude mice.
