本文对无菌培养条件下黄瓜子叶外植体龄对子叶生根能力的影响进行了研究,结果表明,对于分别萌发5天、10天、15天、20天的子叶在MS基本培养基上,无论有无生长素NAA,均以5天龄的子叶生根能力最强。附加10mg/LNAA,显著促进子叶生根。对子叶起始过氧化物酶活性测定表明,子叶起始过氧化物酶活性与生根能力之间存在有一定的关系。
In the present study, the effects of explant age of cucumber on rooting ability of cotyledon in aseptic culture were studied. The results showed that for the cotyledons on MS medium which germinated for 5 days, 10 days, 15 days and 20 days respectively, With or without auxin NAA, all five-day-old cotyledon strongest ability to root. Addition of 1 0 mg / L NAA significantly promoted rooting of cotyledons. The cotyledon initial peroxidase activity assay showed that there is a certain relationship between cotyledon initial peroxidase activity and rooting ability.
