目的:观察骨髓间充质干细胞移植对缺血-再灌注损伤的肠黏膜通透性变化的影响。方法:体外分离、培养和扩增雄性SD大鼠骨髓间充质干细胞,将雌性SD大鼠随机分为三组,A组为假手术组,仅行剖腹,不夹闭肠系膜上动脉;B组大鼠行肠系膜上动脉夹闭45min后开放,取培养至第三代的骨髓间充质干细胞直接肠黏膜下注射移植;C组大鼠夹闭肠系膜上动脉45min后开放,黏膜下注射等量的等渗盐水。术后第3和第6天行乳果糖/甘露醇(L/M)灌胃,6h后收集尿液测定尿L/M比值。取回肠组织行Y染色体原位杂交检测供鼠来源的细胞。测定血浆D-乳酸水平。结果:移植的骨髓间充质干细胞能向缺血-再灌注损伤的肠黏膜归巢,细胞移植的大鼠肠黏膜通透性较C组明显降低(P<0.05)。结论:骨髓间充质干细胞移植可抑制缺血-再灌注损伤后肠黏膜通透性的升高,维护肠黏膜机械屏障的完整性。
Objective: To observe the effect of bone marrow mesenchymal stem cell transplantation on the changes of intestinal mucosal permeability after ischemia-reperfusion injury. METHODS: Bone marrow-derived mesenchymal stem cells from male SD rats were isolated, cultured and expanded in vitro. Female SD rats were randomly divided into three groups. Group A was sham operation group, The rats were exposed to the superior mesenteric artery for 45min and then the bone marrow-derived mesenchymal stem cells of the third generation were transplanted directly into the intestinal mucosa. The rats in the C group were occluded for 45min after the superior mesenteric artery and were injected subcutaneously Isotonic saline. Lactose / mannitol (L / M) were given orally on the 3rd and 6th day after operation. Urine was collected 6 hours later to determine urinary L / M ratio. The gut tissues were harvested for detection of murine-derived cells by Y chromosome in situ hybridization. Plasma D-lactate levels were measured. Results: Transplanted bone marrow mesenchymal stem cells could homing to the intestinal mucosa with ischemia-reperfusion injury. The permeability of the transplanted rat intestinal mucosa was significantly lower than that of the C group (P <0.05). CONCLUSION: Bone marrow mesenchymal stem cells can inhibit the increase of intestinal mucosal permeability after ischemia-reperfusion injury and maintain the integrity of mechanical barrier of intestinal mucosa.
