目的:探讨愈肾颗粒防治肾纤维化大鼠机制研究。方法:将大鼠按照性别、体重随机分为空白组,模型组、愈肾颗粒组、洛丁新组。运用腺嘌呤灌胃造模的方法建立实验性肾纤维化大鼠模型,采用光学显微镜、免疫组织化学方法,观察、检测各组肝细胞生长因子HGF、BMP-7的表达,24h后给药,观察各实验组21天,28天,42天肾组织HGF、BMP-7表达情况的变化。并采用病理图像分析系统,半定量分析HGF、BMP-7含量表达;结果:在28天、42天,洛丁新与愈肾颗粒两个治疗组的表达明显高于模型组同一实相,彼此之间无明显差异,都与模型组均有明显统计学意义,P<0.01或P<0.05;在21天、42天两个治疗组的表达明显高于模型组同一实相,彼此之间无明显差异,都与模型组均有明显统计学意义(P<0.01或P<0.05);结论:愈肾颗粒有效提高HGF、BMP-7蛋白水平的过低表达,从而改善肾纤维化,潜在的延缓了肾纤维化进程。
Objective: To investigate the mechanism of Yushen Granule in prevention and treatment of renal fibrosis in rats. Methods: Rats were randomly divided into blank group, model group, Yushen Granule group and Luodingxin group according to gender and weight. A rat model of renal fibrosis was established by intragastric adenine injection. The expression of hepatocyte growth factor HGF and BMP-7 in each group was observed and detected by light microscopy and immunohistochemistry. Administration was performed 24 hours later. The changes of the expression of HGF and BMP-7 in kidney tissues at 21 days, 28 days, and 42 days in each experimental group were observed. The pathological image analysis system was used to semi-quantitatively analyze the expression of HGF and BMP-7. Results: At 28 days and 42 days, the expressions of Lodinexin and Yushen Granula were significantly higher in the two treatment groups than in the model group. There was no significant difference between the two groups, both of which had significant statistical significance with the model group, P<0.01 or P<0.05. The expressions of the two treatment groups at the 21st and the 42nd days were significantly higher than that of the model group, and there was no difference between the two groups. Significant differences were statistically significant with the model group (P<0.01 or P<0.05); Conclusion: Yushen Granules can effectively increase the expression of HGF and BMP-7 protein levels, thereby improving renal fibrosis. Delayed the progression of renal fibrosis.
