目的探讨PTTG的表达在非小细胞肺癌(NSCLC)发生、发展中的作用及其与PTEN和Ki67蛋白表达的关系。方法应用免疫组化SP法检测PTTG、PTEN和Ki67三种蛋白在60例NSCLC和19例正常支气管黏膜上皮组织中的表达。结果NSCLC组织中PTTG蛋白表达的阳性率为73.33%(44/60),显著高于正常支气管黏膜上皮组织中的表达(P=0.000);PTTG的表达与肿瘤细胞的分化程度和有无淋巴结转移显著相关,但与患者的年龄、性别及肿瘤的组织学类型无关;NSCLC组织中PTTG表达与PTEN表达无相关关系,而与Ki67表达呈显著正相关。结论NSCLC中PTTG高表达,PTEN低表达,PTTG和Ki67在NSCLC的发生和发展中有协同作用,三者可作为反映NSCLC生物学行为的指标。PTTG将有望成为新的肿瘤分子标记与基因治疗的靶点。
Objective To investigate the role of PTTG expression in the development and progression of non-small cell lung cancer (NSCLC) and its relationship with the expression of PTEN and Ki67. Methods Immunohistochemical SP method was used to detect the expression of PTTG, PTEN and Ki67 in 60 cases of NSCLC and 19 cases of normal bronchial epithelial tissue. Results The positive rate of PTTG protein expression in NSCLC tissues was 73.33% (44/60), which was significantly higher than that in normal bronchial mucosa epithelial tissues (P = 0.000). The expression of PTTG was correlated with the differentiation of tumor cells and lymph node metastasis But not with age, sex and tumor histological type. There was no correlation between PTTG expression and PTEN expression in NSCLC tissues, but positively correlated with Ki67 expression. Conclusion The high expression of PTTG, the low expression of PTEN, and the combination of PTTG and Ki67 in NSCLC have synergistic effects on the occurrence and development of NSCLC. All three can be used as an index to reflect the biological behavior of NSCLC. PTTG will hopefully become the new target of tumor molecular marker and gene therapy.
