目的建立以β-环糊精(β-cyclodextrin,β-CD)和离子液体1-丁基-3-甲基咪唑-L-乳酸盐(1-bu-tyl-3-methylimidazoliumL(+)lactic acid,BMIM-LA)联合作为手性选择剂,用于分离扑尔敏、卡维地洛和文拉法辛对映体的毛细管电泳方法。方法采用未涂层熔融石英毛细管柱,背景电解质为Tris-HCl缓冲溶液,运行电压20 kV。考察了手性选择剂β-CD和离子液体的浓度、pH值、缓冲溶液浓度对对映体分离的影响。结果在最优化的电泳条件下,扑尔敏可达到完全分离,卡维地洛和文拉法辛部分分离,分离度分别为2.1、1.2和0.75。结论β-CD与离子液体联用可提高扑尔敏、卡维地洛和文拉法辛对映体的分离选择性。
OBJECTIVE To establish a method for the determination of 1-bu-tyl-3-methylimidazolium (L-lactic acid) in the presence of β-cyclodextrin acid, BMIM-LA) as a chiral selector for the enantiomeric separation of chlorpheniramine, carvedilol and venlafaxine by capillary electrophoresis. Methods The uncoated fused silica capillary column was used. The background electrolyte was Tris-HCl buffer and the voltage was 20 kV. The effects of chiral selectors β-CD and ionic liquid concentration, pH and buffer concentration on enantiomeric separation were investigated. Results Under optimal electrophoresis conditions, chlorpheniramine was completely separated and carvedilol and venlafaxine were partially separated with resolution of 2.1, 1.2 and 0.75, respectively. Conclusion The combination of β-CD with ionic liquid can improve the separation selectivity of enantiomers of chlorpheniramine, carvedilol and venlafaxine.
