目的建立一种快速测定茶鲜叶中β-胡萝卜素的高效液相色谱(HPLC)方法。方法以丙酮为提取溶剂;色谱柱为SunFire~(TM)C_(18)(150 mm×4.6 mm,5μm);以三氯甲烷-乙腈溶液(9:1,V:V)为流动相,等梯度洗脱;波长450 nm;流速1.0 mL/min;柱温35℃。结果在该色谱条件下β-胡萝卜素与其他物质得到了有效的分离,且β-胡萝卜素在9.5×10~(-3)~0.76μg范围内与峰面积有良好的线性关系(r=0.99997),回收率97.32%~101.26%。结论该方法重复性好、精密度高、回收率较好,可作为茶鲜叶中β-胡萝卜素的定量测定方法。
Objective To establish a rapid HPLC method for the determination of β-carotene in tea leaves. Methods The extraction solvent was acetone. The column was SunFire C18 (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of chloroform and acetonitrile (9: 1, V: V) Gradient elution; wavelength 450 nm; flow rate 1.0 mL / min; column temperature 35 ℃. Results The results showed that β-carotene was separated from other substances under the chromatographic conditions, and there was a good linear relationship between β-carotene and peak area in the range of 9.5 × 10 -3 ~ 0.76 μg (r = 0.99997 ), The recovery rate of 97.32% ~ 101.26%. Conclusion The method has good repeatability, high precision and good recovery. It can be used as a quantitative method for the determination of β-carotene in fresh tea leaves.
