目的探讨激素敏感性脂肪酶(HSL)基因(LIPE)启动子C-60G多态性在高糖膳食诱导的健康青年血浆糖脂代谢指标变化中的作用。方法广告招募56名平均年龄为(22.89±1.80)岁的在校健康志愿者(男性27名,女性29名),给予7 d平衡膳食(31%脂肪、54%碳水化合物、15%蛋白质)和6 d高糖膳食(15%脂肪、70%碳水化合物和15%蛋白质),于第1、8、14 d取12 h空腹静脉血,测定血脂、血糖、胰岛素水平,并计算体重指数(BMI)及稳态模型估算胰岛素抵抗指数(HOMA-IR)值。提取基因组DNA,聚合酶链反应-限制性片段长度多态性法分析LIPEC-60G多态性。结果不论在高糖膳食前还是高糖膳食后,CC基因型女性高密度脂蛋白胆固醇(HDL-C)(P<0.01或P<0.05)和载脂蛋白A-Ⅰ(apo A-Ⅰ)(P<0.01或P<0.001)较CC基因型男性升高;高糖膳食后,CC基因型女性总胆固醇(TC)(P<0.01)、低密度脂蛋白胆固醇(LDL-C)(P<0.05)较CC基因型男性升高。与高糖膳食前相比,高糖膳食后CC基因型男性BMI(P<0.05)、TC(P<0.001)和LDL-C(P<0.01)降低,HDL-C(P<0.01)和apo A-Ⅰ(P<0.05)升高;CG基因型男性BMI(P<0.05)和TC(P<0.01)降低。CC基因型女性TC(P<0.001)、LDL-C(P<0.01)降低,甘油三酯(TG)(P<0.01)和胰岛素(P<0.05)升高;CG基因型女性TC降低(P<0.05),TG升高(P<0.05)。结论 LIPEC-60G变异能够抑制高糖膳食诱导的健康青年男性LDL-C降低及HDL-C和apoA-Ⅰ的升高,抑制高糖诱导的健康青年女性LDL-C降低和胰岛素升高。
Objective To investigate the role of the C-60G polymorphism of the hormone-sensitive lipase (LIPE) promoter in the changes of plasma glucose and lipid profile induced by high-glucose diet in healthy youth. Methods Fifty-six healthy volunteers (27 males and 29 females) with a mean age of 22.89 ± 1.80 years old were enrolled in this study. Seven-week balanced diet (31% fat, 54% carbohydrates and 15% protein) 6 d high glucose diet (15% fat, 70% carbohydrates and 15% protein), fasting venous blood was collected on the 1st, 8th, 14th day for determination of blood lipids, blood glucose and insulin, and body mass index (BMI) And homeostasis model assessment of insulin resistance index (HOMA-IR) values. The genomic DNA was extracted and the LIPEC-60G polymorphism was analyzed by polymerase chain reaction-restriction fragment length polymorphism. Results Compared with HDL-C (P <0.01 or P <0.05) and apo A-Ⅰ (CC) genotypes, (P <0.01 or P <0.001) than those with CC genotype. After high glucose diet, the total cholesterol (TC) and the low density lipoprotein cholesterol (LDL-C) ) Than men with CC genotype increased. Compared with those before high glucose diet, CC genotype had significantly lower BMI (P <0.05), TC (P <0.001) and LDL-C (P <0.01), HDL-C A-Ⅰ (P <0.05). The CG genotype of male BMI (P <0.05) and TC (P <0.01) decreased. CC genotype female TC (P <0.001), LDL-C (P <0.01), triglyceride (TG) (P <0.01) and insulin (P < <0.05), TG increased (P <0.05). Conclusion LIPEC-60G mutation can inhibit LDL-C and HDL-C and apoA-I in healthy young men induced by high-glucose diet and inhibit LDL-C and insulin in healthy high-glucose-induced young women.
