在30个不同的玉米材料中,均诱导出愈伤组织,其中15个材料分化出单倍体苗。接种花粉发育时期为单核中期的花药,单倍体愈伤组织(或胚状体)诱导频率最高可达3.25%,比接种花粉为单核靠边期的花药提高3.92倍。诱导愈伤组织的培养基为N_6+2,4-D 2毫克/升+K 1毫克/升+CH 500毫克/升。分化培养基为 N_6+K 1毫克/升+CH 500毫克/升。通过花粉压片的显微镜观察,已见到2—4个细胞的花粉粒和多细胞团。植株根尖压片,染色体数为10,证明是来源于花粉的单倍体植株。单倍体植株已移入土中,并用0.025—0.05%的秋水仙碱溶液进行加倍处理,植株生长正常,最早的植株已抽雄吐丝。
Callus was induced in 30 different maize materials, 15 of which differentiated haploid seedlings. Inoculation of anther at the monocyte stage during pollen development showed that the induction frequency of haploid callus (or embryoid body) was up to 3.25%, which was 3.92 times higher than that of the anther on the monolayer. The medium for callus induction was N_6 + 2,4-D 2 mg / L + K 1 mg / L + CH 500 mg / L. Differentiation medium N_6 + K 1 mg / L + CH 500 mg / L. By microscopic observation of pollen tablets, pollen grains and multicellular aggregates of 2-4 cells have been seen. Plant root tip tablet, chromosome number 10, proved to be derived from pollen haploid plants. Haploid plants have been moved into the soil and doubled with 0.025-0.05% colchicine solution. The plants grew normally and the earliest plants had sprouted silk.
