【目的】观察活血化瘀中药蒲黄提取物蒲黄总黄酮(PTF)对2型糖尿病大鼠炎症因子和胰岛素敏感性的影响。【方法】选用SD大鼠,采用高脂饮食+小剂量链脲佐菌素诱导2型糖尿病大鼠模型,随机分为模型组、PTF组(剂量为200 mg·kg-1·d-1)和罗格列酮组(剂量为4 mg·kg-1·d-1),并设立对照组。干预治疗4周后,检测血浆白细胞介素(IL-6)、肿瘤坏死因子-α(TNF-α),并行胰岛素耐量试验,分析骨骼肌组织细胞因子信号转导抑制因子-3(SOCS-3)蛋白。【结果】治疗4周后,与对照组比较,模型组大鼠血浆IL-6、TNF-α和胰岛素抵抗指数(HOMA-IR)以及骨骼肌组织SOCS-3水平均显著升高(P﹤0.05),胰岛素耐量试验受损(P﹤0.05);与模型组比较,PTF组大鼠血浆IL-6和HOMA-IR水平均显著降低(P﹤0.05),胰岛素耐量试验受损得到显著改善(P﹤0.05),骨骼肌组织SOCS-3水平均显著低于模型组和罗格列酮组(P﹤0.05)。【结论】PTF能降低2型糖尿病大鼠血浆IL-6和骨骼肌组织SOCS-3水平,改善其胰岛素敏感性。
【Objective】 To observe the effects of Puhuang extract of Puhuang extract (PTF) on inflammatory cytokines and insulin sensitivity in type 2 diabetic rats. 【Methods】 SD rats were used to induce type 2 diabetic rats induced by high-fat diet + low-dose streptozotocin (STZ). The rats were randomly divided into model group, PTF group (200 mg · kg-1 · d-1) And rosiglitazone (4 mg · kg-1 · d-1), and the control group was established. After intervention for 4 weeks, serum interleukin (IL) -6, tumor necrosis factor-α (TNF-α) and parallel insulin tolerance test were tested. The levels of cytosolic signaling inhibitor- )protein. 【Results】 After treatment for 4 weeks, the levels of plasma IL-6, TNF-α, insulin resistance index (HOMA-IR) and skeletal muscle SOCS-3 in model group were significantly higher than those in control group (P <0.05). Compared with the model group, the levels of plasma IL-6 and HOMA-IR in the PTF group were significantly decreased (P <0.05), and the impaired insulin resistance test was significantly improved (P <0.05). SOCS-3 levels in skeletal muscle were significantly lower than those in model group and rosiglitazone group (P <0.05). 【Conclusion】 PTF can reduce the levels of plasma IL-6 and skeletal muscle tissue SOCS-3 in type 2 diabetic rats and improve their insulin sensitivity.
