目的:探讨缺血预处理(IPC)对实验性大鼠缺血再灌注(IR)心肌梗死(MI)面积及过氧化物酶体增殖物激活受体α(PPARα)表达的影响。方法:Wistar大鼠70只随机分为3组:对照组(24只),IR组(24只),IPC组(22只)。采用鼠心冠状动脉左前降支结扎法制作体内IR模型,观察MI范围,运用半定量RT-PCR方法对心肌PPARαmRNA的表达情况进行分析,运用Western-blotting方法检测PPARα蛋白表达情况。结果:IPC组梗死面积显著减少(P<0·05),并且心肌PPARαmRNA及蛋白表达水平显著上调(P<0·05)。结论:IPC可显著减少实验性大鼠IR MI面积及并使心肌中PPARα显著上调。
Objective: To investigate the effect of ischemic preconditioning (IPC) on myocardial infarct (MI) area and peroxisome proliferator - activated receptor α (PPARα) expression in experimental rats with ischemia - reperfusion (IR). Methods: Seventy Wistar rats were randomly divided into three groups: control group (24 rats), IR group (24 rats) and IPC group (22 rats). In vivo IR model was established by ligation of left anterior descending coronary artery of rat heart. MI range was observed. The expression of PPARα mRNA in myocardium was analyzed by semi-quantitative RT-PCR. The protein expression of PPARα was detected by Western-blotting. Results: The area of infarction in IPC group was significantly decreased (P <0.05), and the mRNA and protein expression of PPARα in myocardium were significantly increased (P <0.05). Conclusion: IPC can significantly reduce the area of IR MI in experimental rats and significantly up-regulate PPARα in myocardium.
