目的:加味香砂六君子汤在胃癌前病变(PLGC)模型大鼠胃黏膜的Bax、Bcl-2、P53mRNA表达方面的影响及作用机理研究。方法:120只健康Wistar大鼠按照随机数字表法分为空白对照组和模型组、维霉素组、加味香砂六君子汤高、中及低剂量组,造模后给予药物干预治疗,通过免疫组化法,检测Bcl-2、Bax及P53mRNA表达水平。结果:加味香砂六君子汤各剂量组Bcl-2蛋白阳性表达率较模型组及维霉素组显著降低(P<0.05),各剂量组Bax蛋白阳性表达率较模型组及维霉素组明显增高(P<0.05)。各干预组p53mRNA蛋白表达不同程度降低,且明显低于模型组(P<0.05);其中加味香砂六君子汤中剂量组的作用效果最显著,与其他各组相比,差异显著(P<0.05)。结论:加味香砂六君子汤可调控胃癌前期病变大鼠胃黏膜Bcl-2、Bax表达水平,降低p53mRNA的表达,具有治疗PLGC的作用。
Objective: To study the effect of Xiangsha Liujunzi Decoction on gastric mucosal expression of Bax, Bcl-2, P53mRNA in gastric precancerous lesions (PLGC) model rats and its mechanism of action. Methods: One hundred and twenty healthy Wistar rats were divided into blank control group, model group, imipenem group, Jiaxiang Xiangsha Liujunzi Decoction high, medium and low dose groups according to random number table. Immunohistochemistry was used to detect the expression of Bcl-2, Bax and P53 mRNA. Results: The positive rate of Bcl-2 protein expression in each dose group of Modified Xiangsha Liujunzi Decoction was significantly lower than that of the model group and the doxycycline group (P <0.05). The positive rate of Bax protein in each dose group was significantly higher than that of the model group and the doxycycline group Was significantly higher (P <0.05). The expression of p53 mRNA in each intervention group decreased to different extents, which was significantly lower than that in model group (P <0.05). The effect of Jiaxiang Xiangsha Liujunzi Decoction was the most significant, with significant difference compared with other groups (P < 0.05). Conclusion: Jiaxiang Xiangsha Liujunzi Decoction can regulate the expression of Bcl-2 and Bax in gastric mucosa and reduce the expression of p53 mRNA in the gastric precancerous lesions, which has the therapeutic effect on PLGC.
