目的:检测EGFR外显子19缺失突变(以下简称EGFR Del 19)和信号分子JAK/STAT在NSCLC组织中的表达,探讨EGFR Del 19表达与下游信号通路JAK/STAT表达之间的相关性,为进一步研究EGFR Del 19之NSCLC发生发展机制提供依据。方法:经ARMS法筛选出125例EGFR Del 19的NSCLC组织,应用免疫组织化学法检测该组织样本中EGFR Del 19、p-JAK1和p-STAT1的表达,统计分析三者在临床病理特点中的表达差异及EGFR Del 19与p-JAK1、p-STAT1表达之间的相关性。结果:EGFR Del 19阳性主要在细胞膜上表达,p-JAK1、p-STAT1阳性表达主要定位于细胞质和细胞核中。EGFR Del 19与NSCLC的病理分级、TNM分期、淋巴结转移与否显著相关(P值均<0.05)。EGFR Del 19与p-JAK1、p-STAT1表达在不同病理分级、TNM分期及淋巴结转移与否组间具有显著相关性(P值均<0.05,rs值均>0.3)。结论:EGFR Del 19可能通过活化JAK1-STAT1途径促进NSCLC的发生发展。
OBJECTIVE: To detect the expression of EGFR Del 19 mutation and JAK / STAT signaling in NSCLC and to investigate the correlation between EGFR Del 19 expression and downstream JAK / STAT signaling To further study the EGFR Del 19 NSCLC provide the basis for the mechanism of development. Methods: 125 cases of EGFR Del 19 NSCLC were screened by ARMS. The expression of EGFR Del 19, p-JAK1 and p-STAT1 in the tissue samples were detected by immunohistochemical method. Statistical analysis was made on the three clinical and pathological features Expression differences and EGFR Del 19 p-JAK1, p-STAT1 expression between the correlation. Results: The positive expression of EGFR Del 19 was mainly expressed on the cell membrane. The positive expression of p-JAK1 and p-STAT1 mainly localized in the cytoplasm and nucleus. EGFR Del 19 was significantly associated with pathological grade, TNM stage and lymph node metastasis (all P <0.05). The expression of EGFR Del 19 and p-JAK1, p-STAT1 in different pathological grade, TNM stage and lymph node metastasis were significantly correlated (P <0.05, rs> 0.3). Conclusion: EGFR Del 19 may promote the development of NSCLC through activation of JAK1-STAT1 pathway.
