本实验通过肌肉和侧脑室注射五肽胃泌素(G_5),初步探讨其对大鼠心肌细胞动作电位幅值(APA)及心率(HR)的影响。结果如下:1)侧脑室内注射G_510μg/10μl时,对APA及HR无明显影响;当注射剂量加倍(20μg/10μl)时,在注射30min才使HR显著变慢(P<0.01),而APA仍无明显变化。2)肌注G_560μg/100g,使HR显著变慢,APA显著降低。事先肌注阿托品 25mg/100g后,再注G_5,上述效应明显减弱或消失(P<0.01)。以上结果表明,G_5对心脏电活动具有抑制作用,该作用主要是通过外周胆碱能系统而实现的。
In this study, pentagastrin (G_5) was injected into muscle and lateral ventricle to investigate the effects of pentagastrin on cardiomyocyte action potential amplitude (APA) and heart rate (HR). The results were as follows: 1) When intracerebroventricular injection of G_510μg / 10μl did not have obvious effect on APA and HR; when the injection dose was doubled (20μg / 10μl), HR was significantly slowed down 30min after injection (P <0.01) Still no significant change. 2) intramuscular G_560μg / 100g, HR significantly slowed, APA significantly lower. Prior to intramuscular injection of atropine 25mg / 100g, then note G_5, the above effect was significantly weakened or disappeared (P <0.01). The above results show that, G_5 has an inhibitory effect on cardiac electrical activity, which is mainly achieved by the peripheral cholinergic system.
