目的:采用血清药理学方法研究绿谷灵芝对低密度脂蛋白(LDL)氧化作用的影响,以及对氧化型低密度脂蛋白(ox-LDL)和晚期糖基化终末产物(AGE)诱导的单核细胞内皮细胞粘附作用的影响。方法:通过检测培养上清中硫代巴比妥酸反应物质(TBARS)的生成表示含药血清对LDL的氧化作用,用白细胞髓过氧化物酶法测定单核细胞对内皮细胞的粘附作用。结果:0.12g/kg绿谷灵芝一次给药组0.5h、1h、2h、3h,两次给药0.5h、1h取血分离得到的血清对LDL氧化作用无明显影响,两次给药2h、3h以及0.12g/kg、0.24g/kg、0.72g/kg绿谷灵芝连续给药10天后,取血分离得到的血清均可降低LDL的氧化作用(P<0.05);0.24g/kg、0.72g/kg绿谷灵芝连续给药10天后,取血分离得到的血清均可抑制由ox-LDL和AGE诱导的单核细胞对内皮细胞的粘附作用(P<0.05)。结论:绿谷灵芝含药血清能显著降低LDL的氧化作用,减轻由ox-LDL和AGE诱导的单核细胞对内皮细胞的粘附作用。
OBJECTIVE: To investigate the effect of L. lucidum on the oxidation of low-density lipoprotein (LDL) using serum pharmacology and the induction of oxidized low-density lipoprotein (ox-LDL) and advanced glycation end products (AGE). Effects of Monocyte Endothelial Cell Adhesion. METHODS: The oxidative effect of drug-containing serum on LDL was measured by measuring the production of thiobarbituric acid-reactive substance (TBARS) in the culture supernatant, and the adhesion of monocytes to endothelial cells was measured by leukocyte myeloperoxidase method. . RESULTS: 0.12g/kg Lugu Ganoderma lucidum was administered for 0.5h, 1h, 2h, and 3h in one administration group. Blood serum obtained after two administrations of 0.5h and 1h had no significant effect on the oxidation of LDL. After 3h and 0.12g/kg, 0.24g/kg, 0.72g/kg Lugu Ganoderma lucidum was continuously administered for 10 days, blood serum obtained could reduce the oxidation of LDL (P<0.05); 0.24g/kg, 0.72 After 10 days of continuous administration of g/kg Lugu Ganoderma lucidum, serum obtained from blood sampling could inhibit the adhesion of mononuclear cells to endothelial cells induced by ox-LDL and AGE (P<0.05). Conclusion: Lugu Ganoderma drug-containing serum can significantly reduce the oxidation of LDL and reduce the adhesion of monocytes induced by ox-LDL and AGE to endothelial cells.
