目的:建立快速测定鼻炎喷剂中盐酸麻黄碱含量的分析方法。方法:采用傅里叶近红外(FT-NIR)液体透射光谱法采集样品的近红外光谱。以高效液相色谱法测得值为参考,运用偏最小二乘法,以一阶微分(1d)+Norris平滑为预处理方法,谱段选择6136.38~5364.99 cm-1和7 038.90~6 969.48cm-1建立定量校正模型。结果:定量模型校正集的R2为0.992 6,校正集验证均方差(RMSEC)为1.20;验证集的R2为0.993 5,外部验证均方差(RMSEP)为1.28,内部交叉验证R2为0.986 9,内部交叉验证均方差(RMSECV)为1.60。结论:所建立的定量模型具有满意的拟合效果和预测能力,能快速检测鼻炎喷剂中盐酸麻黄碱的含量,可推广应用于生产过程中的在线分析。
Objective: To establish a rapid method for the determination of ephedrine hydrochloride in rhinitis spray. Methods: The near-infrared spectra of the samples were collected by FT-NIR liquid transmission spectroscopy. Using the method of partial least squares (1d) + Norris smoothing as the pretreatment method, the data of 6136.38 ~ 5364.99 cm-1 and 7 038.90 ~ 6 969.48 cm- 1 Establish a quantitative calibration model. Results: The R2 of the calibration set of the quantitative model was 0.992 6, the RMSEC of the calibration set was 1.20, the R2 of the validation set was 0.993 5, the root mean square error of external validation (RMSEP) was 1.28, the internal cross-validation R2 was 0.986 9, The cross-validation mean square error (RMSECV) was 1.60. Conclusion: The established quantitative model has a satisfactory fitting effect and predictive ability, can quickly detect the content of ephedrine hydrochloride in rhinitis spray, and can be widely applied to the on-line analysis in the production process.
