目的探究十子代平方的10种组方药物以及每种药物的主要成分对α-葡萄糖苷酶活性的影响,明确该方对α-葡萄糖苷酶起抑制作用的主要成分。方法利用蔗糖为底物的体外α-葡萄糖苷酶筛选模型,以葡萄糖氧化酶法测定10种中药及主要成分对α-葡萄糖苷酶的抑制率;以阿卡波糖为阳性对照,分别计算各实验组的抑制率和半数抑制率(IC50)。结果 10种中药及10种单体成分均有一定α-葡萄糖苷酶抑制作用。其中,菟丝子、茺蔚子、五味子、莱菔子、紫苏子5种中药和迷迭香酸、金丝桃苷、芥子碱硫氰酸盐3种成分的抑制作用较强,呈明显的浓度-效应关系。当浓度为125 mg/m L时,紫苏子、菟丝子、五味子、莱菔子、茺蔚子的抑制率分别为58.49%、55.32%、54.01%、53.13%和50.67%,同浓度阿卡波糖的抑制率为92.09%;当浓度为40 mmol/L时,迷迭香酸、金丝桃苷、芥子碱硫氰酸盐的抑制率分别为93.52%、87.10%、78.11%,同浓度阿卡波糖的抑制率为90.89%。菟丝子、茺蔚子、五味子、莱菔子、紫苏子、迷迭香酸、金丝桃苷、芥子碱硫氰酸盐和阿卡波糖对α-葡萄糖苷酶抑制作用的IC50分别为44.401、89.608、72.146、82.139、21.549、0.412、0.605、0.860和0.376 mg/m L。结论 10种中药及10种单体成分均可一定程度上抑制α-葡萄糖苷酶,中药菟丝子、茺蔚子、五味子、莱菔子、紫苏子和单体迷迭香酸、金丝桃苷、芥子碱硫氰酸盐对α-葡萄糖苷酶的抑制力较强。
