目的:考察不同产地的生地黄中所含地黄寡糖和梓醇的含量。方法:采用水为溶媒,超声提取糖类成分,流动相用乙腈-水(72∶28),流速1.0 mL·min-1,NH2色谱柱分离,示差折光检测器检测,高效液相色谱法测定;采用甲醇为溶媒,超声提取梓醇,流动相用甲醇-0.1%磷酸溶液(1∶99),流速1.0 mL·min-1,C18色谱柱分离,检测波长210 nm,高效液相色谱法测定。结果:产地不同,生地黄中的地黄寡糖成分、含量不同,梓醇含量不同。结论:本方法操作简便、准确、快速、干扰少,适用于生地黄中地黄寡糖和梓醇的含量测定。
Objective: To investigate the content of Rehmannia oligosaccharides and sterols in Rehmanniae collected from different areas. METHODS: Water was used as solvent, ultrasonic extraction of carbohydrates, mobile phase with acetonitrile-water (72:28), flow rate 1.0 mL·min-1, NH2 column separation, differential scanning detector detection, high performance liquid chromatography The methanol was used as a solvent, ultrasonic extraction of sterols, mobile phase with methanol-0.1% phosphoric acid solution (1:99), flow rate 1.0 mL · min-1, C18 column separation, detection wavelength 210 nm, HPLC determination . RESULTS: The origin of different habitats, Rehmanniae oligosaccharides in the composition, content of different sterol content. Conclusion: This method is simple, accurate, rapid, and less interfered. It is suitable for determination of oligosaccharides and sterols in Rehmannia glutinosa.
