本文用反相色谱法测定人血浆中的游离氨基酸。氨基酸通过异硫氰酸苯酯(PITC)柱前衍生后形成苯基硫酸盐(PTC)-氨基酸。以流动相A液及B液洗脱,B液梯度洗脱0~49%,UV检测波长254nm,色谱柱温(38±1)℃。该法不仅可用于蛋白质水解氨基酸的分析,而且特别适用于血液等体液中生理氨基酸的定性、定量分析,具有分析时间短,灵敏度高的特点。其相关系数均值为0.9997,变异系数均值为2.42%,回收率为94.83%。
This article uses reverse phase chromatography to determine free amino acids in human plasma. Amino acids were phenylpropionate (PITC) pre-column derivatized to form phenylsulfate (PTC) -amino acids. The mobile phase A and B were eluted. The gradient of B was eluted from 0 to 49%. The UV detection wavelength was 254 nm and the temperature of the column was (38 ± 1) ℃. This method can not only be used for the analysis of amino acids in proteolysis, but also is especially suitable for the qualitative and quantitative analysis of physiological amino acids in body fluid such as blood, which has the advantages of short analysis time and high sensitivity. The mean of correlation coefficient was 0.9997, the mean of coefficient of variation was 2.42% and the recovery rate was 94.83%.
