目的通过对H22荷瘤小鼠肿瘤组织微小核糖核酸(miRNAs)表达谱的检测,探讨消癌解毒方抗肝癌的作用机制。方法将50只H22荷瘤小鼠随机分为模型组,消癌解毒方低剂量组、中剂量组、高剂量组和顺铂组,每组10只。应用病理组织学技术,检测各组H22荷瘤小鼠肿瘤组织光镜下的差异表现。应用miRNA芯片技术,检测各组H22荷瘤小鼠肿瘤组织差异表达miRNAs。应用统计学方法分析,找出与消癌解毒方作用机制相关的差异miRNAs。结果病理组织学检查结果显示,随着消癌解毒方剂量的增加,病理性核分裂像减少,癌细胞减小,抗癌效果增强。根据miRNA芯片的分析结果,消癌解毒方对H22瘤荷小鼠肿瘤组织内一些特异miRNAs如miR-1298-5p、miR-874-3p、miR-721、miR-298-5 p、miR-551 b-5 p、miR-346-5 p、miR-105表达显著上调;miR-24-3 p、miR-3963、miR-127-3 p、miR-434-5 p、miR-1187、miR-468-3 p、miR-221-5 p、miR-6695-5 p表达显著下调。结论消癌解毒方可能通过调控多种miRNAs的表达来发挥其抗肿瘤的作用。
Objective To investigate the mechanism of Xiaojian Jiedu Fang against liver cancer by detecting the expression patterns of miRNAs in tumor tissues of H22 tumor-bearing mice. Methods 50 H22 tumor-bearing mice were randomly divided into model group, Xiaowangjiedu Fang low dose group, middle dose group, high dose group and cisplatin group, with 10 mice in each group. The histopathology was used to detect the difference of tumor tissue in H22 tumor-bearing mice under light microscope. MiRNA microarray was used to detect differentially expressed miRNAs in H22 tumor-bearing mice. Application of statistical methods to identify and find the mechanism of action associated with Xiaojiandudu different miRNAs. Results The results of histopathological examination showed that the pathological mitosis was decreased, the number of cancer cells was decreased and the anti-cancer effect was enhanced with the increase of the dosage of Xiaojian Jiedu. According to the analysis results of miRNA microarray, Xiaojian Jiedu decoction can inhibit specific miRNAs such as miR-1298-5p, miR-874-3p, miR-721, miR-298-5p, miR-551 miR-346-5p and miR-105 were significantly up-regulated in miR-24-3p, miR-363p, miR-434-5p, miR- 468-3 p, miR-221-5 p, miR-6695-5 p expression was significantly down-regulated. Conclusion Xiaojian Jiedu Fang may exert its anti-tumor effect by regulating the expression of various miRNAs.
