目的研究人巨细胞病毒(hum an cytom egalovirus,H C M V)O R F11基因在临床分离株中的多态性及其与H C M V感染所致不同消化系统疾病之间的关系。方法研究对象为16名具有不同消化系统疾病的H C M V感染儿。对全部标本进行H C M V O R F11基因的半巢式PC R扩增。应用D N Aclub,BioEdit,PR O SITE数据库及D N Astar软件对PC R产物的序列进行分析。结果所有临床分离株H C M V O R F11的核苷酸序列是保守的,而其编码蛋白翻译后修饰位点的氨基酸序列则高度保守。未发现H C M V O R F11序列的变异与不同表现类型的消化系统疾病相关。结论 H C M V O R F11基因相对保守,不同分离株的序列变异与不同临床表现的消化系统疾病没有明确的相关性。
Objective To study the polymorphism of human cytomegalovirus (HCM) O R F11 gene in clinical isolates and its relationship with different digestive diseases caused by H C M V infection. METHODS: Sixteen HCCV-infected children with different digestive diseases were studied. All samples were subjected to semi-nested PC R amplification of the H C M V O R F11 gene. The PC R product sequences were analyzed using D N Aclub, BioEdit, PR O SITE databases and D N Astar software. Results The nucleotide sequences of all clinical isolates, H C M V O R F11, were conserved and their amino acid sequences encoding the post-translational modification sites were highly conserved. No variation in the H C M V O R F11 sequence was found to be associated with different manifestations of digestive diseases. Conclusion The gene H C M V O R F11 is relatively conservative. There is no clear correlation between the sequence variations of different isolates and different clinical manifestations of digestive diseases.
