目的:研究苦参碱对人肝癌细胞系SMMC-7721的诱导分化作用.方法:用细胞培养方法,观察细胞增殖、形态、功能和代谢等方面指标,研究苦参碱(Ma)对人肝癌细胞系SMMC-7721在体外的诱导分化作用.结果:各处理组细胞均出现细胞增殖抑制,亚细胞结构趋于正常,AFP分泌量明显低于培养天数相同的对照组细胞,γ-GT活力逐日下降,酪氨酸-α-酮戊二酸转移酶(TAT)活力始终高于对照组细胞.处理6d后,细胞DNA含量降低,S期细胞数增加.结论:Ma可使SMMC-7721细胞聚集于S期,诱导其向正常肝细胞分化
Objective: To study the induction and differentiation of matrine on human hepatoma cell line SMMC-7721. Methods: The effects of matrine (Ma) on human hepatocellular carcinoma cell line SMMC-7721 in vitro were studied in vitro by using cell culture method to observe the cell proliferation, morphology, function and metabolism. Results: The cells in all treatment groups showed cell proliferation inhibition, subcellular structure tended to be normal, and the secretion of AFP was significantly lower than that of the control group cells with the same number of days of culture. The activity of γ-GT decreased day by day, and tyrosine-α-ketoglutarate Transaminase (TAT) activity is always higher than the control group cells. After 6 days of treatment, the cellular DNA content decreased and the number of S phase cells increased. Conclusion: Ma can induce SMMC-7721 cells to accumulate in S phase and induce their differentiation into normal hepatocytes
