目的探讨溶血磷脂酸受体1(Lysophosphatidic acid receptor 1,LPAR1)在人脑胶质瘤组织中的表达及意义。方法收集临床切除的经病理证实的良性胶质瘤组织、恶性胶质瘤组织及正常脑组织,采用RT-PCR法及免疫组织化学染色法检测各组脑组织中LPAR1在mRNA及蛋白水平的表达,并分析LPAR1表达与脑胶质瘤临床病理特征之间的相关性。结果正常脑组织、良性胶质瘤组织、恶性胶质瘤组织中均有LPAR1 mRNA及蛋白的表达;胶质瘤组织中LPAR1 mRNA及蛋白的表达水平均明显高于正常脑组织,差异有统计学意义(P<0.05),且恶性胶质瘤组织中LPAR1蛋白的表达水平明显高于良性胶质瘤组织(P<0.05);LPAR1的表达与脑胶质瘤临床病理分级相关,而与年龄、性别、肿瘤部位无关。结论 LPAR1在脑胶质瘤组织中表达增高,可能在胶质瘤发生发展的过程中发挥重要作用。
Objective To investigate the expression of Lysophosphatidic acid receptor 1 (LPAR1) in human glioma and its significance. Methods The clinically resected specimens of benign glioma, malignant glioma and normal brain tissues were collected. The mRNA and protein levels of LPAR1 in each group were detected by RT-PCR and immunohistochemical staining , And analyzed the correlation between LPAR1 expression and clinicopathological characteristics of glioma. Results The expression of LPAR1 mRNA and protein in normal brain tissue, benign glioma and glioma tissues were both higher than that in normal brain tissue. The expression of LPAR1 mRNA and protein in glioma tissues were significantly higher than that in normal brain tissues (P <0.05), and the expression of LPAR1 protein in malignant glioma tissues was significantly higher than that in benign glioma tissues (P <0.05). The expression of LPAR1 correlated with the clinicopathological grade of glioma, but not with age, Sex, tumor site has nothing to do. Conclusion LPAR1 is highly expressed in glioma tissues and may play an important role in the development of glioma.
