细胞内蛋白 蛋白的结合是丙型肝炎病毒 (HCV)与宿主肝细胞相互作用的分子生物学基础。HCV的非结构蛋白 5A(NS5A)被认为是一种HCV基因组编码的重要调节蛋白因子 ,参与HCV的复制、转录和信号传导等过程 ,但是它在HCV的致病及耐药等方面的作用还存有争议。为了进一步阐明这个多功能蛋白质与宿主蛋白的关系 ,作者采用酵母双杂交系统 3,在酵母细胞融合蛋白表达型人肝cDNA文库中进行筛选 ,得到 35个与NS5A特异性结合的阳性克隆 ,包括载脂蛋白、线粒体单体型基因、磷脂酸酸性磷酸酶等 11种已知功能蛋白质基因和 3个未知功能基因。本研究结果为阐明NS5A在HCV致病中的作用提供了重要研究线索
Binding of intracellular protein proteins is the molecular biological basis for the interaction of Hepatitis C virus (HCV) with host hepatocytes. The nonstructural protein 5A (NS5A) of HCV is considered as an important regulatory protein factor encoded by the HCV genome and involved in the process of HCV replication, transcription and signal transduction, but its role in the pathogenesis and drug resistance of HCV also Controversial. To further elucidate the relationship between this multifunctional protein and host protein, the authors used yeast two-hybrid system 3 to screen yeast-cell fusion protein-expressing human liver cDNA library to obtain 35 positive clones that specifically bind to NS5A, Lipoprotein, mitochondrial haplotype, phospholipid acid phosphatase and other 11 kinds of known function protein genes and 3 unknown function genes. The results of this study provide important clues to clarify the role of NS5A in the pathogenesis of HCV
