目的观察liguzinediol(2,5-二羟甲基-3,6-二甲基吡嗪)对正常SD大鼠心脏血流动力学的影响。方法将大鼠随机分为5组:生理盐水组,阳性对照组(磷酸二酯酶抑制剂米力农,0.3mg·kg-1),3个剂量组(liguzinediol5,10,20mg·kg-1),每组10只。采用RM6240B/C(四道)型多道生理记录仪测量给药后大鼠心脏血流动力学指标如左心室收缩压(LVSP),左心室终末舒张压(LVEDP),左心室最大变化速率(±dp/dtmax),左室开始收缩至左室内压上升速率峰值时间(t-dp/dtmax),收缩压(SAP),舒张压(DAP),平均压(MAP),脉压差(MPP)以及心率(HR)。结果静脉给药后,liguzinediol5,10,20mg·kg-1使大鼠左心室LVSP、+dp/dtmax、-dp/dtmax、SAP、MAP、MPP显著增加,t-dp/dtmax、LVEDP降低。结论liguzinediol对正常大鼠心脏具有正性肌力作用。
Objective To investigate the effects of liguzinediol (2,5- dimethylol-3,6-dimethylpyrazine) on the hemodynamics of normal SD rats. Methods The rats were randomly divided into 5 groups: normal saline group, positive control group (milrinone 0.3 mg · kg -1), 3 doses (liguzinediol 5, 10, 20 mg · kg -1 ), 10 in each group. The hemodynamic indexes of rats such as left ventricular systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), maximum left ventricular rate of change (± dp / dtmax), left ventricular systolic pressure (t-dp / dtmax), systolic pressure (SAP), diastolic pressure (DAP), mean arterial pressure (MAP) ) And heart rate (HR). Results After intravenous administration of liguzinediol, 10, 20 mg · kg-1, LVSP, + dp / dtmax, -dp / dtmax, SAP, MAP and MPP were significantly increased and t-dp / dtmax and LVEDP were decreased. Conclusion liguzinediol has a positive inotropic effect on the heart of normal rats.
