目的 :在宫内发育迟缓 (IUGR)模型的基础上 ,测定小于胎龄 (SGA)新生幼鼠肝脏组织胰岛素样生长因子 -Ⅰ(IGF -Ⅰ )及其结合蛋白 (IGFBPs)的基因表达变化 ,以阐明它们在发病中的作用。方法 :子宫动脉结扎 (UAL)建立IUGR模型 ,用逆转录 (RT) -PCR分析SGA新生幼鼠肝脏组织IGF -Ⅰ、IGFBPsmRNA表达变化。结果 :轻度SGA肝脏组织IGF -ⅠmRNA水平无明显改变 ,IGFBP1 mRNA水平升高 ,重度SGA同时伴有IGF -ⅠmRNA水平下降 ,IGFBP1 、IGFBP2 mRNA水平上升。结论 :肝脏组织IGF系统基因表达异常可能是胎儿宫内生长发育迟缓的重要病因之一。
OBJECTIVE: To determine the gene expression of insulin-like growth factor-I (IGF-I) and its binding protein (IGFBPs) in the liver of young immature young (SGA) rats based on IUGR model. To clarify their role in the pathogenesis. Methods: IUGR model was established by uterine artery ligation (UAL). The expression of IGF-I and IGFBPs mRNA in the liver of SGA newborn rats were detected by RT-PCR. Results: There was no significant change in IGF-ⅠmRNA level in mild SGA liver tissue, IGFBP1 mRNA level was increased in severe SGA liver, accompanied by decreased IGF-ⅠmRNA and IGFBP1, IGFBP2 mRNA levels in severe SGA. Conclusion: Abnormal gene expression of IGF in liver tissue may be one of the important causes of fetal growth retardation.
