目的:建立同时测定大卫颗粒中绿原酸、连翘苷、黄芩苷的高效液相色谱法。方法:采用Sinochrom ODS-BP(250mm×4.6mm,5μm)色谱柱,流动相为甲醇:30mmol·L~(-1)磷酸二氢钾溶液,梯度洗脱,流速为1.0ml·min~(-1),检测波长为277nm。结果:绿原酸在10~80μg·ml~(-1)范围内线性关系良好(r=0.9996),平均回收率为98.85%,RSD=1.02%(n=9);连翘苷在10~80μg·ml~(-1)范围内线性关系良好(r=0.9993),平均回收率为99.17%,RSD=0.99%(n=9);黄芩苷50~600μg·ml~(-1)范围内线性关系良好(r=0.9995),平均回收率为98.72%,RSD=1.10%(n=9)。结论:本方法简便易行,能够准确、快速测定大卫颗粒中的绿原酸、连翘苷和黄芩苷的含量。
Objective: To establish a simultaneous determination of chlorogenic acid, forsythin, baicalin in David particles by high performance liquid chromatography. Methods: The chromatographic column of Sinochrom ODS-BP (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of methanol: 30 mmol·L -1 potassium dihydrogen phosphate solution with gradient elution at a flow rate of 1.0 ml · min ~ 1), detection wavelength of 277nm. Results: The linear range of chlorogenic acid was 10 ~ 80μg · ml ~ (-1) (r = 0.9996), the average recovery was 98.85% and the RSD was 1.02% (n = 9) The average recovery was 99.17% and RSD was 0.99% (n = 9) in the range of 80μg · ml ~ (-1), and the linear range was 50 ~ 600μg · ml ~ The linear relationship was good (r = 0.9995). The average recovery was 98.72% with RSD = 1.10% (n = 9). Conclusion: The method is simple and easy to determine the content of chlorogenic acid, forsythin and baicalin in David granules accurately and rapidly.
