目的:研究狼毒大戟配伍大枣对肝硬化腹水大鼠模型的抵抗作用。方法:采用苯巴比妥联合四氯化碳(CCl4)法建立肝硬化腹水大鼠模型,将大鼠随机分为正常组、肝硬化腹水模型组、狼毒大戟组,狼毒大戟配伍大枣组,每组各10只。观测大鼠水代谢及苏木素-伊红(HE)染色观察肝组织病理变化,用免疫组织化学法测定I型胶原(CollagenⅠ),Ⅲ型胶原(CollagenⅢ),纤维粘连蛋白(Fibronectin,FN),层粘连蛋白(Laminin,LN),结蛋白(Desmin)在肝组织中的表达。结果:与模型组比较,狼毒大戟组及狼毒大戟配伍大枣组大鼠24 h尿量明显增多(P<0.01),腹腔积液显著减少(P<0.01)。给狼毒大戟配伍大枣组大鼠肝脏纤维间隔及变性程度较模型组有明显改善。免疫组化结果显示,肝硬化模型组及狼毒大戟治疗组,Ⅰ,Ⅲ型胶原及纤维连接蛋白的含量均显著高于正常组(P<0.01),模型组LN和Desmin的含量显著高于正常组(P<0.01),狼毒大戟配伍大枣组能显著降低LN和Desmin的积分吸光度IA。结论:狼毒大戟配伍大枣可明显改善肝硬化腹水大鼠肝硬化病理进程:增加尿量,促进腹水排泄,其作用机制可能与狼毒大戟配伍大枣对CollagenⅠ,CollagenⅢ,FN,LN,Desmin的调控有关。
Objective: To study the effect of Radix Euphorbiae combined with jujube on the rat model of cirrhosis induced by ascites. Methods: The rat model of cirrhosis induced by ascites was established by phenobarbital combined with carbon tetrachloride (CCl4). The rats were randomly divided into normal group, cirrhosis model group, Euphorbia pulmonarum group, Jujube group, each group of 10. The pathological changes of liver tissue were observed under water metabolism and hematoxylin-eosin (HE) staining. The expressions of collagenⅠ, collagen Ⅲ, fibronectin (FN) Laminin (LN), Desmin in liver tissue. Results: Compared with the model group, the urine volume of rats of Radix Euphorbiae and Radix Euphorbia combined with jujube group increased significantly (P <0.01) and the amount of ascites decreased significantly (P <0.01). To the Euphorbia Euphorbia jujube group liver fibrosis and degeneration degree than the model group has significantly improved. The results of immunohistochemistry showed that the content of typeⅠ, type Ⅲ collagen and fibronectin in liver cirrhosis model group and Euphorbia pulcherrima group were significantly higher than those in normal group (P <0.01), and the contents of LN and Desmin in model group were significantly higher In the normal group (P <0.01), Euphorbia Euphorica compatibility jujube group can significantly reduce the integral absorbance of LN and Desmin IA. Conclusion: Radix Euphorbia combined with jujube can significantly improve the cirrhosis of rats with cirrhosis of liver cirrhosis pathological process: increased urine output and promote the excretion of ascites, the mechanism may be compatibility Euphorbia jujuba Collagen Ⅰ, Collagen Ⅲ, FN, LN , Desmin’s regulation.
