脑缺血时中缝背核5-HT变化的免疫组化观察

目的:探讨沙土鼠脑缺血时中缝背核神经元5-HT的变化。方法:用免疫组织化学ABC法及图象分析对沙土鼠脑缺血不同时间以及缺血10分钟后再灌不同时间其中缝背核神经元5-HT的变化进行研究。结果:脑缺血10分钟、30分钟时,沙土鼠脑中缝背核5-HT阳性神经元的平均光密度值(OD值)与对照组比较无显著性差异;而缺血4小时时则明显降低,差异有显著性(P<0.05);脑缺血10分钟后,再灌流30分钟,5-HT下降不明显,而4小时时则明显降低(P<0.05),至24小时时达最低水平(P<0.05),36小时逐渐恢复正常。结论:当缺血至一定时间或缺血10分钟再灌流到一定时间时,中缝背核神经元内5-HT的含量减少。 Objective: To investigate the changes of 5-HT in dorsal raphe nucleus during gerbil cerebral ischemia. Methods: The changes of 5-HT in dorsal raphe nucleus neurons were detected by immunohistochemical ABC method and image analysis at different time points after gerbil ischemia and 10 minutes after ischemia. Results: The mean optical density (OD) of 5-HT positive neurons in dorsal raphe nucleus of gerbils at 10 minutes and 30 minutes after cerebral ischemia had no significant difference compared with that of the control group, but obvious at 4 hours after ischemia (P <0.05). After 10 minutes of cerebral ischemia, the decrease of 5-HT was not obvious at 30 minutes after reperfusion, but significantly lower at 4 hours (P <0.05), and lowest at 24 hours Level (P <0.05), gradually returned to normal after 36 hours. CONCLUSIONS: The content of 5-HT in the dorsal raphe nucleus neurons decreased when ischemia for a certain period of time or reperfusion for 10 minutes after ischemia.
