用离体实验法研究ι─荷包牡丹碱对豚鼠结肠带收缩的影响,结果发现:ι─荷包牡丹碱明显拮抗组织胺及ca2+引起的收缩,呈现非竞争性拮抗方式,pD'2分别为4.92及5.43.在无ca2+液中,ι─荷包牡丹碱与罂粟碱相似,能抑制组织胺诱发的结肠带收缩.提示两药对结肠带依赖细胞内Ca2+所致的收缩均有明显的抑制,当恢复细胞外Ca2+浓度后,两药对依赖细胞外Ca2+经ROC内流所致的收缩无影响.
The effect of ι-bicuculline on contractile activity of guinea pig colon strips was investigated by in vitro experiments. The results showed that ι-bicuculline significantly antagonized the contraction induced by histamine and ca2 +, showing a noncompetitive antagonistic manner with pD’2 values of 4 .92 and 5.43. In ca2 + -free solution, ibuprofen is similar to papaverine and inhibits histamine-induced contractions of the colon. These results suggest that both drugs inhibit the contractility induced by intracellular Ca2 + in colonic strips obviously. When the extracellular Ca2 + concentration is restored, the two drugs have no effect on the contraction of extracellular Ca2 + dependent on influx of ROC.
