以15个不同产地的黄连(味连,CoptisChinensisFranch)药材为试验材料,应用RAPD分子标记技术对其种质资源的遗传多样性进行了分析。20条10merRAPD引物共得到148条扩增条带,其中有39条呈现多态性,占26.4%,说明黄连的遗传多样性水平较低。用TreeconW软件分析遗传距离,变化范围在0.039~0.108之间,表明黄连种质间的亲缘关系较近。UPGMA聚类分析显示黄连种质的地理界限并不明显,但还是可以看出来源于同一地区的部分黄连种质聚在一起。
The genetic diversity of the germplasm resources was analyzed using RAPD molecular markers in 15 different habitats of Coptis (Coptis Chinensis Franch). A total of 148 amplified bands were obtained from 20 10mer RAPD primers, 39 of which were polymorphic, accounting for 26.4%, indicating that the genetic diversity of Coptis chinensis was low. Using TreeconW software to analyze the genetic distance, the variation ranged from 0.039 to 0.108, indicating that the genetic relationship between the Huanglian germplasm was closer. UPGMA cluster analysis showed that the geographic boundaries of Coptis germplasm are not obvious, but it can be seen that part of the Coptis germplasm originating from the same area is clustered together.
