目的探讨依布硒啉对内毒素性急性肺损伤(ALI)的保护作用。方法将48只健康雄性SD大鼠随机分为6组:正常对照组、模型组、依布硒啉低剂量组、依布硒啉中剂量组、依布硒啉高剂量组和地塞米松组,每组8只。大鼠尾静脉注射脂多糖5.0mg/kg建立ALI模型,依布硒啉低剂量组、依布硒啉中剂量组、依布硒啉高剂量组和地塞米松组大鼠分别于腹腔注射依布硒啉7.5、15.0、30.0mg/kg及地塞米松5.0mg/kg,正常对照组和模型组大鼠分别注入等量溶剂。处理后6h,麻醉放血处死大鼠,收集支气管肺泡灌洗液(BALF)检测蛋白质浓度,采用酶联免疫吸附测定(ELISA)法检测肺组织匀浆中肿瘤坏死因子α(TNF-α)和白细胞介素1β(IL-1β)的浓度。光镜下观察肺组织病理学改变。结果依布硒啉治疗组大鼠BALF中蛋白质浓度及肺组织中TNF-α和IL-1β浓度明显低于与模型组(P<0.05);肺组织病理学表现有明显改善。结论依布硒啉对内毒素性ALI有一定保护作用,其机制与抑制炎症因子有关。
Objective To investigate the protective effect of ebselen on endotoxin-induced acute lung injury (ALI). Methods Forty-eight healthy male Sprague-Dawley rats were randomly divided into 6 groups: normal control group, model group, Ebselen low dose group, Ebselen medium dose group, Ebselen high dose group and Dexamethasone group , Each group of 8. Rats were injected with lipopolysaccharide (5.0 mg / kg) via the tail vein to establish ALI model. The rats in low dose Ebselen group, medium dose Ebselen group, high dose Ebselen dose group and dexamethasone group were injected intraperitoneally Cloth selenium 7.5, 15.0, 30.0mg / kg and dexamethasone 5.0mg / kg, the normal control group and model group rats were injected with the same amount of solvent. Six hours after the treatment, rats were killed by anesthesia, bronchoalveolar lavage fluid (BALF) was collected to detect the protein concentration, and the levels of tumor necrosis factor-α (TNF-α) and leukocytes in the lung homogenate were measured by enzyme linked immunosorbent assay (ELISA) The concentration of Interleukin 1β (IL-1β). Lung histopathological changes were observed under light microscope. Results The concentration of protein in BALF and the concentrations of TNF-α and IL-1β in lung tissue of Ebselen-treated rats were significantly lower than those of model rats (P <0.05). The pathological changes of lungs were improved obviously. Conclusion Ebselen has a protective effect on endotoxin-induced ALI, and its mechanism is related to the inhibition of inflammatory cytokines.
