目的探讨子宫腺肌病中程序性死亡因子-1(PD-1)及其配体-1(PD-L1)的表达,以及PD-1/PD-L1信号通路在子宫腺肌病发生发展中的作用。方法采用免疫组织化学法及Western blotting检测不同月经周期的子宫腺肌病在位内膜与异位子宫内膜、正常子宫内膜组织标本中PD-1、PD-L1蛋白的表达(每组增生期:n=15、分泌期:n=15)。分析不同组别间PD-1、PD-L1蛋白的表达差异及其与月经周期的关系。结果免疫组织化学结果显示,PD-1、PD-L1在不同月经周期的正常子宫内膜及腺肌病在位、异位子宫内膜中均有表达。腺肌病异位子宫内膜中PD-1蛋白(P<0.01)与PD-L1蛋白(P<0.01)表达升高。Western blotting结果显示,腺肌病在位子宫内膜中PD-1蛋白(P<0.01)与PD-L1蛋白(P<0.01)的表达增高。但在腺肌病组内不同月经周期中,PD-1/PD-L1的表达水平差异无统计学意义(P=0.12;P=0.45)。结论 PD-1蛋白及PD-L1蛋白在腺肌病在位及异位子宫内膜中的表达升高,提示PD-1/PD-L1信号通路可能参与子宫腺肌病的发生、发展过程。
Objective To investigate the expression of PD-1 and PD-L1 in adenomyosis and the role of PD-1 / PD-L1 signaling in the development of adenomyosis Role. Methods Immunohistochemistry and Western blotting were used to detect the expression of PD-1 and PD-L1 protein in eutopic and ectopic endometrium and eutopic endometrium of different menstrual cycles Period: n = 15, Secretory phase: n = 15). The differences of PD-1 and PD-L1 protein expression between different groups and their relationship with menstrual cycle were analyzed. Results The results of immunohistochemistry showed that PD-1 and PD-L1 were expressed in both normal endometrium and adenomyosis at different menstrual cycles in ectopic endometrium. The expression of PD-1 protein (P <0.01) and PD-L1 protein (P <0.01) in ectopic endometrium of adenomyosis increased. Western blotting showed that the expression of PD-1 protein (P <0.01) and PD-L1 protein (P <0.01) in endometriosis of adenomyosis increased. However, there was no significant difference in PD-1 / PD-L1 expression between different menstrual cycles in adenomyosis group (P = 0.12; P = 0.45). Conclusions PD-1 protein and PD-L1 protein are highly expressed in eutopic and ectopic endometrium of adenomyosis, suggesting that PD-1 / PD-L1 signaling pathway may be involved in the development and progression of adenomyosis.
