目的探讨垂体瘤转化基凶(PTTG)及与之密切相关的碱性成纤维生长因子(bFGF)在胶质瘤组织中的表达规律,探讨其与肿瘤增殖、肿瘤血管生成、临床病理之间的关系。方法用RT-PCR分析了32例胶质瘤组织和6例正常脑组织标本PTTG mRNA的表达;用SP法检测bFGF、PCNA及CD34在肿瘤中的表达,计算微血管密度(MVD),并结合临床病理资料进行分析。结果胶质瘤组织中PTTG基因呈过度表达,高级别(Ⅲ~Ⅳ级)胶质瘤组织中PTTG mRNA的平均表达水平(0.91±0.34)显著高于低级别(Ⅰ~Ⅱ级)胶质瘤(0.47±0.33)及正常脑组织(0.11±0.17),3组间分别比较,差异均有显著性意义(P<0.05);生存时间1≥2年组中PTTG mRNA的表达水平(0.52±0.39)低于生存时间<2年组(0.97±0.26),两者比较差异有显著性意义(P=0.001);低级别和高级别胶质瘤组的bFGF、PCNA及MVD相比均有显著差异性(P<0.05);PTTG mRNA和MVD、PCNA、bFGF之间存在显著正相关;与患者性别、年龄、肿瘤部位、肿瘤大小均无相关关系。结论PTTG参与胶质瘤的早期发生。bFGF则对血管形成、肿瘤增殖有明显作用,二者均和肿瘤的侵袭性密切相关。
Objective To investigate the expression of PTTG and its closely related basic fibroblast growth factor (bFGF) in glioma tissues, and to explore its relationship with tumor proliferation, tumor angiogenesis and clinical pathology relationship. Methods The expression of PTTG mRNA in 32 glioma tissues and 6 normal brain tissue samples was analyzed by RT-PCR. The expression of bFGF, PCNA and CD34 in tumor was detected by SP method, and the microvessel density (MVD) Pathological data were analyzed. Results The PTTG gene was overexpressed in glioma tissues. The average expression level of PTTG mRNA (0.91 ± 0.34) in high grade glioma tissues (Ⅲ ~ Ⅳ grade) was significantly higher than that in low grade ) Glioma (0.47 ± 0.33) and normal brain tissue (0.11 ± 0.17) respectively. There were significant differences between the three groups (P <0.05) The expression level of PTTG mRNA in the 2-year group (0.52 ± 0.39) was lower than that in the 2-year survival group (0.97 ± 0.26) (P = 0. 001). The levels of bFGF, PCNA and MVD in low-grade and high-grade gliomas were significantly different (P <0.05). There was a significant positive correlation between PTTG mRNA, MVD, PCNA and bFGF. Gender, age, tumor location, tumor size were not related. Conclusions PTTG is involved in the early stage of glioma. bFGF has a significant effect on angiogenesis and tumor proliferation, both of which are closely related to tumor aggressiveness.
