目的 研究血小板衍化生长因子 B(PDGF)基因转染成纤维细胞后对其生长增殖的影响。方法 构建PDGF B真核表达质粒 ,用脂质体LipofectAMINE介导转染成纤维细胞。相差显微镜观察成纤维细胞生长状况 ,MTT法测绘成纤维细胞生长曲线。结果 PDGF B基因转染的成纤维细胞生长良好 ,细胞生长曲线显示PDGF B基因转染 3d后的成纤维细胞较未转染的成纤维细胞生长明显增快 (P <0 0 5 )。结论 PDGF B基因转染后可显著加速成纤维细胞的生长增殖。
Objective To study the effect of platelet-derived growth factor B (PDGF) gene on the proliferation of fibroblasts after transfection. Methods The eukaryotic expression plasmid of PDGF B was constructed and transfected into fibroblasts by LipofectAMINE. Fibroblast growth was observed by phase contrast microscopy and fibroblast growth curve was plotted by MTT assay. Results The fibroblasts transfected with PDGF B gene grew well. The cell growth curve showed that the growth of fibroblasts after PDGF B gene transfection was significantly faster than that of untransfected fibroblasts (P <0.05). Conclusion PDGF B gene transfection can significantly accelerate the growth and proliferation of fibroblasts.
