本文报告了利用-氯化碘法对人血清低密度脂蛋白(LDL)进行~(125)Ⅰ碘化标记,结果表明,~(125)Ⅰ-LDL的比放射性为(61~185)×10~(-6)μCi/ng LDL,脂质标记率<1%,游离~(125)Ⅰ含量在0.43~1.90%之间,~(125)Ⅰ-LDL浓度为0.5~1.0mg/ml.~(125)Ⅰ-LDL的生物活性经受体实验证实与天然脂蛋白无差异,基本符合用于脂蛋白代谢研究的要求.本法具有反应温和,不易损伤脂蛋白,脂质标记率低,设备简单等优点,为进一步进行脂蛋白受体的研究提供了可行的方法.
In this paper, 125I iodinated labeling of human serum low density lipoprotein (LDL) by iodinated iodine method was reported. The results showed that the specific radioactivity of ~ (125) Ⅰ-LDL was (61 ~ 185) × 10 ~ (-6) μCi / ng LDL, the lipid labeling rate was less than 1%, the content of free 125 I was between 0.43 and 1.90%, the concentration of 125 I-LDL was 0.5 ~ 1.0 mg / (125) The biological activity of Ⅰ-LDL showed no difference with that of natural lipoprotein, which was basically in line with the requirements for the study of lipoprotein metabolism.This method has the advantages of mild reaction, low damage to lipoprotein, low lipid labeling rate, Simple and so on, for the further study of lipoprotein receptor provides a viable method.
