目的研究早期宫颈鳞癌差异表达基因整合素α2、白细胞介素-8(IL-8)和间隙连接蛋白-43(Cx43)在宫颈鳞癌中的表达及其与淋巴结转移的关系,探讨其在宫颈鳞癌淋巴结转移中的作用机制。方法采用反转录-聚合酶链反应(RT-PCR)、组织芯片进行的免疫组织化学方法检测早期宫颈鳞癌组织中整合素α2、IL-8和Cx43mRNA及蛋白的表达。结果有淋巴结转移的早期宫颈鳞癌组织中整合素α2和IL-8的表达显著高于非转移组织(P<0.05),CX43的表达则相反(P<0.01)。且整合素α2和IL-8的表达呈正相关(r=0.241,P<0.05)。结论整合素α2、IL-8和CX43与早期宫颈鳞癌的淋巴结转移密切相关,在一定程度上发挥了促进和抑制早期宫颈鳞癌淋巴结转移的作用。
Objective To study the expression of integrin α2, interleukin-8 (IL-8) and connexin-43 (Cx43) in cervical squamous cell carcinoma and their relationship with lymph node metastasis in early stage of cervical squamous cell carcinoma Mechanism of lymph node metastasis in cervical squamous cell carcinoma. Methods The expression of integrin α2, IL-8 and Cx43 mRNA and protein in early stage of cervical squamous cell carcinoma were detected by reverse transcription-polymerase chain reaction (RT-PCR) and tissue microarray immunohistochemistry. Results The expression of integrin α2 and IL-8 in early cervical squamous cell carcinoma with lymph node metastasis was significantly higher than that in non-metastatic tissue (P <0.05) and CX43 (P <0.01). There was a positive correlation between integrin α2 and IL-8 (r = 0.241, P <0.05). Conclusion Integrin α2, IL-8 and CX43 are closely related to lymph node metastasis in early stage of cervical squamous cell carcinoma and play a role in promoting and inhibiting lymph node metastasis of early stage cervical squamous cell carcinoma to a certain extent.
