抗日本血吸虫可溶性虫卵抗原IgY的制备及初步应用

目的制备抗日本血吸虫可溶性虫卵抗原(Soluble egg antigen,SEA)鸡卵黄免疫球蛋白(Egg yolk immunoglobulin,Ig Y),探讨其应用于血吸虫病流行区查病的可行性。方法在血吸虫病流行区采集间接红细胞凝集试验(IHA)阳性者尿液,在非流行区采集健康人尿液。以SEA经皮下注射免疫莱杭母鸡获得anti-SEA Ig Y,以十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)测定其相对分子量;并以anti-SEA Ig Y作为捕捉抗体,采用双抗体夹心法ELISA检测IHA阳性者和健康人尿液中的血吸虫循环可溶性虫卵抗原(Circulating soluble egg antigen,CSEA)。结果成功制备并纯化antiSEA Ig Y。共检测IHA阳性者尿液48份,在尿液中检出CSEA阳性26例,检出率为54.17%;检测健康人尿液10份,均为阴性。结论基于Ig Y的免疫诊断技术可有效检出人尿液中CSEA,其作为一种便捷、无创伤的新方法可应用于血吸虫病查病工作中。 Objective To prepare Egg yolk immunoglobulin (Ig Y) against soluble egg antigen (SEA) of Schistosoma japonicum and investigate the feasibility of its application in the endemic area of ​​schistosomiasis. Methods Urine was collected by indirect hemagglutination test (IHA) in the endemic area of ​​schistosomiasis, and healthy human urine was collected in the non-endemic area. Anti-SEA Ig Y was immunized with SEA by subcutaneous injection, and the relative molecular weight was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Anti-SEA Ig Y Antibodies were captured and the circulating soluble egg antigen (CSEA) of Schistosoma japonicum in IHA positive and healthy human urine was detected by double antibody sandwich ELISA. Results AntiSEA Ig Y was successfully prepared and purified. A total of 48 urine samples were tested positive for IHA, 26 samples were positive for CSEA in urine and the detection rate was 54.17%. 10 urine samples from healthy people were negative. Conclusion IgY immunodiagnosis technique can effectively detect CSEA in human urine. As a convenient and noninvasive method, CSEA can be applied to the diagnosis of schistosomiasis.
