目的分析铁离子应答元件结合蛋白1基因在不同中华按蚊品系中表达量的差异。方法根据前期二维电泳筛选获得的差异表达肽段序列,设计相应的PCR引物扩增基因片段,并检测中华按蚊抗性品系、敏感品系、现场采集品系铁离子应答元件结合蛋白1基因表达水平的差异。结果扩增获得了中华按蚊铁离子应答元件结合蛋白1的基因片段,经荧光定量PCR证实,该基因在抗性品系蚊体内的表达量是敏感品系的9.42倍,现场品系是敏感品系的6.38倍。结论中华按蚊铁离子应答元件结合蛋白1基因在杀虫剂抗性通路中有一定的作用,可做为新的抗性治理靶标及基因检测指标。
OBJECTIVE: To analyze the difference of the expression of iron-binding element-binding protein 1 gene in different Anopheles sinensis strains. Methods According to the differentially expressed peptide sequences obtained from the 2D electrophoresis screening in the early stage, the corresponding PCR primers were designed to amplify the gene fragments and detect the expression level of Fe-I receptor binding protein 1 gene in the resistant and susceptible strains of Anopheles sinensis The difference.
